CORONA Insights: 2021: Project Updates and Promising Treatments

2021 was a productive year for the CORONA Project. Through the efforts of many volunteers and staff at the CSTL, we expanded the database to include data from all known randomized controlled trials conducted in Covid-19 patients. This broad inclusion of all RCTs allowed the CORONA Project to identify promising  treatments early in the research process, before findings are confirmed by large-scale clinical trials. With this expanded dataset, which includes information on the size of the trial, primary endpoint, p-values, and effect sizes, we developed an algorithm to objectively review the published data to determine an aggregate estimate of effectiveness. By removing subjective analysis and interpretation, these grades and rankings are independent of external preferences and biases that may mislead patients and clinicians looking to determine the most effective treatments.

Updated algorithm

The updated STORM algorithm depends on several datapoints that are collected from published RCTs. The primary endpoint of each trial is evaluated to determine if the outcomes indicate that the treatment is beneficial or not. The p-values for each primary endpoint are aggregated by a formula to determine the likelihood that at least one trial has a significant result.

Transparency and free access to data are core goals of the CORONA Project. As we updated and expanded our dataset, the public viewer was updated to accommodate the growing body of evidence. In addition to grades for each drug, the STORM viewer now allows a user to see all the extracted data from any published paper, which can be downloaded for further analysis. Grades have been expanded to include separate grades for prioritization for research, and clinical efficacy.
